Abstract: In this talk we will discuss the current panorama on Chagas disease drug discovery and the bottlenecks that are believed to have prevented more substantial advances towards new drugs in the past years. A still superficial understanding of the disease pathogenesis and the lack of screening assays and animal models that can correctly reproduce essential features of the disease in both its acute and chronic stages are probably some of underlying reasons for the recent failure of posaconazole and ravuconazole in Phase II trials for Chagas disease treatment. By combing High Content Screening with a Chemical Genetics approach, and taking into consideration the genetic and phenotypic heterogeneity of Trypanosoma cruzi, our work has focused in the development of novel and physiologically relevant assays that might be able to predict with greater confidence the therapeutic potential of new drug candidates and result in lower attrition rates due to lack of efficacy.
