Tropical neglected diseases are a group of diseases that affect underprivileged populations and are characterized by the lack of pharmaceutical R&D investments. In this group, leishmaniasis and trypanosomiases are responsible for more than 80 thousand deaths annually and over 10 million cases worldwide. Current treatment options had been developed decades ago and are invariably associated with low efficacy, high toxicity, high costs and/or low treatment compliance due to difficult administration regimens. In recent years, the development and implementation of systems for high throughput screening has helped to reduce the innovation gap in drug discovery. In this seminar will be discussed the strategy for drug discovery adopted by our group, which has been developing phenotypic assays based on cellular models of these diseases. HCS assays were used to screen small molecule compound libraries obtained from Pharma, totaling over 2 million screening points and resulting in hits representative of several new chemical classes. The secondary assays developed are used in the preliminary characterization of the mechanism of action of the compounds, and are becoming essential in the process of the hit and lead candidate prioritization. Currently we are expanding our portfolio of secondary assays, with emphasis on Chagas disease, to establish PK/PD relationships in vitro and to correlate the data obtained with data from in vivo studies in patients and in experimental animal models, thus establishing a set of assays and skills able to predict with greater confidence the therapeutic potential of new drug candidates.
