Proteins are prodigal molecules in binding to other chemical entities, either organic or inorganic; these interactions are mostly related to the diversity of organic radicals neatly distributed in the space by thermodynamicallyfolding of the polypeptide chain.
This spatial organization allows either the binding to small molecules or macromolecules, including other proteins, DNA, RNA and polysaccharides.
The determination of macromolecule complex structures at high-resolution is a big challenge for structural biology, as they are often flexible and transient molecules to be crystallized or too large to be solved by nuclear magnetic resonance.
Thus, the creation of accurate theoretical structural models that allow the understanding of macromolecular interactions contribute to the elucidation of complex biological processes, allowing to guide bench experiments, as well as to feed back the models with experimental data, improving their predictive power.
In this seminar, it will be presented the current stage of the researches developed in the Bioinformatics Facility (LBI) of LNBio, aiming at the creation and the use of computational methods to build structural models of biomacromolecular complexes. The seminar will comprise discussions about the aspects of reduction of spatial complexity, scoring functions and the use of experimental data as crystallographic data, chemical cross-linking and exchange of hydrogen/ deuterium obtained by mass spectrometry and known ligands to increase the usability of these models in the development of modulados of the protein activity, as well as the understanding of molecular signaling mechanisms mediated by biomacromolecular interactions.
