Abstract: The Citrus sinensis cyclophilin CsCyp is a target of the Xanthomonas citri TAL effector PthA, required to elicit tumors on citrus. CsCyp binds the C-terminal domain of the citrus RNA Polymerase II, affecting transcription of PthA-regulated genes. CsCyp belongs to the subfamily of divergent cyclophilins, which are characterized by an additional loop, the divergent loop, two invariable cysteines, and a conserved glutamate. Despite the suggested roles in ATP and metal binding, the function of these unique structural elements remained unknown for many years. In this seminar, we will show how divergent cyclophilins regulate their peptidyl-prolyl cis-trans isomearse activity. Our data reveal a new type of allosteric regulation in divergent cyclophilins, involving disulfide bond formation and a loop displacement mechanism.
