Abstract: The Thyroid Hormone Receptors (TRs) are members of the nuclear receptors superfamily, which act as transcription regulators modulated by ligand binding. The regulation of these proteins by thyroid hormones (T3 and T4) has been related to cell development, differentiation and proliferation; carbohydrate metabolism, oxygen consumption; and renal, cardiac and central nervous system functions. The most common pathologies related to these receptors are hypo and hyperthyroidism, metabolic syndrome and cancer. In this context, these receptors are interesting targets for new drugs development.
Given the importance and the lack of details regarding the mechanism of TRs action, our group intends to seek information about the structural details of these receptors and their interactions on the cellular context, in order to understand the pathways regulated by TRs, which have not yet been described.
In this seminar, it will be presented some results obtained during the last year. Firstly, we will discuss preliminary data on the TRα interactome, highlighting some differences on the pathways regulated by thyroid hormones. Then, we will address more specific issues about the TRs mechanisms of action. Our results have indicated that the TNFα gene promoter is negatively regulated by the thyroid hormones and that this regulation occurs by direct interaction between the receptor and the promoter, which explains, in part, the relation between decreased T3 levels and the triggering of the inflammatory process. Finally, it will be shown the results that demonstrate details on the interaction of TRβ with coactivators, important not only for elucidating the TR mechanism of action, but also for mapping other possible modulation sites.
