Abstract: Bioluminescence is one of the important tools for analysis of biomolecular dynamics. Particularly, the reporter gene assay utilizing luciferin-luciferase reaction is widely used for monitoring gene expression. Conventional reporter gene assay is just a method to measure the transcriptional activity in the end-point, although the activity is changing with time. In recent years, efforts are attempting to apply reporter gene assays to monitor living cells.
We developed a device for real-time reporter gene assay in living cells. This device can continuously measure bioluminescence from culturing cells in an incubator. Therefore this system can be applied to monitor continuously the time course of gene expression level after drug stimulation in living cells. For understanding gene expression networks, it is important to detect the expressions of multiple genes in a same sample and at the same time. We have developed a multi reporter assay system to detect luciferases eliciting bioluminescence of different colors, for application in real-time reporter gene assays.
Bioluminescence imaging analysis is effective because it is able to visualize detailed real-time changes of gene expression at the level of individual cells. To enable imaging analysis with bioluminescence, we developed higher sensitive bioluminescent single cell imaging system. This system is effective to quantitatively trace biomolecular dynamics of single cell level for long term.
