Abstract: Discovery-based proteomics has long been the source of potential candidates for biomarkers and therapeutic targets. However, the list of candidates without follow up is very disappointing. The bottleneck of discovery strategies is also associated to limit ability to use the unbiased and robust statistical methods treating the large-scale data in conjunction, instead of small number of proteins to indicate candidate biomarkers and therapeutic targets. In this scenario, I will discuss about some strategies that have been used in our group to overcome the critical bridge between discovery MS and targeted MS to further validation and clinical implementation.
