Seminar: Regulation of retinoic acid signaling in vertebrate development

Abstract: Retinoic acid (RA) signaling is crucial for correct embryonic development. Raldh2 (Aldh1a2) is the major enzyme involved in retinoic acid synthesis during early development and its expression pattern is dynamic. Little is known about the regulation of Raldh2 expression, despite its expression patterns and functions during development are the focus of many studies. I will show our efforts to unveil cis and trans elements that control Raldh2 expression and describe a new lineage of transgenic mouse, which activates LacZ under the control of an intronic enhancer of the Raldh2 gene. We have demonstrated that this intronic enhancer activates endogenous Raldh2 expression in the dorsal spinal cord, I will also exhibit the functional role of the transcription factor TGIF as a repressor of enhancer activity, placing TGIF as a novel regulator of retinoic acid signaling, through Raldh2 expression, in the developing dorsal spinal cord.
