Proposal submissions will take place from\u00a0<strong>September 1 to October 2, 2026<\/strong>, exclusively through the\u00a0<a href=\"https:\/\/sauonline.cnpem.br\/duo\/\"><strong>SAU Online<\/strong>\u00a0<\/a>system.<\/p>\n<p class=\"isSelectedEnd\" style=\"text-align: justify;\"><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter size-full wp-image-65496\" src=\"https:\/\/i0.wp.com\/cnpem.br\/wp-content\/uploads\/2026\/07\/LEG3.jpg?resize=500%2C281&amp;ssl=1\" alt=\"\" width=\"500\" height=\"281\" data-recalc-dims=\"1\" \/><\/p>\n<p class=\"isSelectedEnd\" style=\"text-align: justify;\">Operated by the Brazilian Biosciences National Laboratory (LNBio), the facility offers technical and scientific support for the generation of mammalian knockout cells using CRISPR\/Cas9 technology, with an emphasis on strategic experimental planning and the production of genome editing tools.<\/p>\n<p class=\"isSelectedEnd\" style=\"text-align: justify;\">The initiative aims to expand national access to gene editing technologies, fostering technological autonomy and accelerating the development of models for biomedical, pharmacological and biotechnological research.<\/p>\n<p class=\"isSelectedEnd\" style=\"text-align: justify;\"><strong>Main services offered by LEG include:<\/strong><\/p>\n<ul style=\"text-align: justify;\" data-spread=\"true\">\n<li><strong>Experimental design and gene editing strategy development:<\/strong>\u00a0Support in defining the most appropriate approach for CRISPR\/Cas9-mediated gene knockout, including guide RNA selection, validation strategies and analysis of potential off-target effects, limited to up to two genes per proposal;<\/li>\n<li><strong>Production of CRISPR tools:<\/strong>\u00a0Manufacturing of customized molecular components such as sgRNAs, plasmid vectors and ribonucleoprotein (RNP) complexes, according to the defined experimental design;<\/li>\n<li><strong>Remote technical support:<\/strong>\u00a0Remote technical guidance to assist users in applying CRISPR tools in their own laboratories, including recommendations on transfection, clone selection and genotypic validation. Support will be provided in up to two remote meetings, with a maximum duration of two hours each, during the project execution period defined in the call.<\/li>\n<\/ul>\n<p class=\"isSelectedEnd\" style=\"text-align: justify;\">As part of the strategy to broaden national access to the technology, proposals from Brazil\u2019s North, Northeast and Central-West regions will be prioritized. Selected users may also follow, in person at LNBio, the experiments related to CRISPR tool production.<\/p>\n<h2 style=\"text-align: justify;\">Eligibility criteria<\/h2>\n<p class=\"isSelectedEnd\" style=\"text-align: justify;\">Eligible proposals must meet the following criteria:<\/p>\n<ul style=\"text-align: justify;\" data-spread=\"true\">\n<li>Projects aimed at constitutive knockout of up to two target genes in mammalian cell lines;<\/li>\n<li>Proposals involving conditional knockout or knock-in strategies will not be accepted;<\/li>\n<li>Only one proposal per principal investigator will be considered. In the event of multiple submissions, only the first proposal submitted will be evaluated.<\/li>\n<\/ul>\n<h2 style=\"text-align: justify;\">How to submit research proposals<\/h2>\n<p class=\"isSelectedEnd\" style=\"text-align: justify;\">Researchers interested in using the LEG infrastructure must submit their proposals via\u00a0<a href=\"https:\/\/sauonline.cnpem.br\/duo\/\"><strong>SAU Online<\/strong><\/a>, a portal managed by CNPEM\u2019s User Office (EdU).<\/p>\n<p class=\"isSelectedEnd\" style=\"text-align: justify;\">LEG calls for proposals are published semiannually.
